Abstract	The antimicrobial capacity of polymorphonuclear leukocytes (PMN) in the presence of dextran sulfate (DS) and heparin, which are similar to naturally occurring mucosubstance in rabbit PMN granules was investigated. The cytochemical activity of PMN granules in phagocytosing and nonphagocytosing cells with and without DS or heparin were also evaluated. No evidence for uptake of DS or heparin was obtained when either phagocytosing or nonphagocytosing PMN were incubated in the presence of these two mucosubstances. Specifically, they did not affect cytochemical reactivity of acid phosphatase in cytoplasmic granules or phagocytic vacuoles at the ultrastructural level. The sulfated acid mucosubstance did not affect reactivity of cytoplasmic granules for acid phosphatase, alkaline phosphatase, peroxidase, dipeptidyl aminopeptidase I (DAP I) or acid mucosubstance at the light microscopic or ultrastructural level. DAP I activity was demonstrated in the primary granules of PMN for the first time and appeared unaffected by DS. Acid phosphatase was not affected by DS or heparin as determined by biochemical assay, but DAP I, II, and III and cathepsin BI were inhibited by DS. The inhibition of the proteases by acid mucopolysaccharide suggests the latter function in PMN granules by keeping proteases in a latent state. The antimicrobial capacity of PMN was shown by viability plate 'count to be inhibited by DS. This could not be attributed to lysosomal enzyme inhibition and appeared in part to result from cytotoxic effects of DS. Cell aggregation and cell injury with presumed loss of phagocytic capacity were apparent from light microscopy and from transmission and scanning electron microscopy. Early myelocytes were shown ultrastructurally to phagocytize and digest gram negative and gram positive bacteria. A full complement of primary and secondary granules in PMN was not necessary for the phagocytic process.
