Abstract	Due to the difficulties in deriving and solving nonlinear rate equations, enzyme kineticists often restrict their steady-state initial velocity studies to simple linear models. However, many enzymatic reactions yield nonlinear kinetic equations, especially over a wide range of substrate concentrations. One method of explaining these nonlinear initial velocity data is by means of random binding mechanisms. This thesis deals with a digital computer modeling and simulation of random binding mechanisms. Based on the topological data of the enzymatic mechanism, the rate equation is automatically derived. Constraint equations arrived from the principle of microreversibility are also generated. A stepwise nonlinear regression technique then obtains a set of rate constants. This derived model is statistically tested for acceptance or rejection. With such a computer system, complex enzymatic mechanisms can be further investigated.
