Abstract	Lymphocytes were separated by the plasmagel method, the Ficol1 Hypaque method, the simple centrifugation method, and the semiautomatic method from blood drawn from ten normal individuals, ten cancer patients, and ten tubercu- 1osis patients. The lymphocyte mixture obtained after each method was analyzed for percent recovery, amount of contamination by erythrocytes and neutrophils, and percent viability. In addition, normal individuals' and tuberculosis patients' lymphocytes were analyzed by a phytomitogen study as a functional test of viability and to show the immunocompetence of the· tuberculosis patients on rifampin, an antituberculosis drug. The results show the Ficoll Hypaque method as the method of choice in normal. individuals, whereas the simple centrifugation method is the method of choice for the cancer and tuberculosis patients. Generally, the best method for all three types of individuals considering percent recovery, least contamination, and optimal viability is the Ficoll Hypaque method The phytomitogen study, done as an ancillary assessment of the viability of Ficol1 Hypaque separated lymphocytes from normal individuals and tuberculosis patients, showed that the rifampin-treated tuberculosis patients seemingly suffered no impairment of cellular mediated immunity as measured by PHA.
