Abstract	Lectins are compounds which bind to terminal and in some cases internal sugars or sugar sequences of oligosaccharides comprising complex carbohydrates. Conjugation of lectins to horseradish peroxidase produces a complex that once bound to complementary glycoconjugates in tissue sections can be subsequently visualized following treatment with the chromagen ,3,3'-diaminobenzidene. Accordingly, the distribution of lectin-reactive glycoconjugates were characterized in normal human kidney as well as in several cases of nephroblastoma (Wilms' tumor). In normal kidney, proximal tubules exhibited a relatively uniform localization of glycoconjugates having bi- and/or triantennary N-linked oligosaccharides, terminal ?-fucose and terminal ?-galactose occurring in virtually all cells. In contrast, collecting ducts and the more distal segments of nephrons showed an extremely heterogeneous distribution of lectin-reactive complex carbohydrates, namely glycogen and those having terminal ?-galactose, ?-galactose, ?-fucose and ?-N-acetylgalactosamine. These compounds occurred in some cells but not others lining thin limbs, distal tubules or collecting ducts. The content and distribution of glycoconjugates in the nephron varied only slightly between kidneys from different individuals but individual variability was extensive in collecting ducts. Lectin staining of seven nephroblastomas revealed similarities between blastema and stroma with respect to their content of glycoconjugates whereas blastema and epithelial cells exhibited major differences. In general, blastema and stroma contained glycoconjugates with terminal or penultimate ?-galactose, glycoconjugates having bi-and/or triantennary N-linked sugar chains, sialoglycoconjugates, and occasionally glycogen. Epithelial cells also showed these complex carbohydrates but stained additionally for terminal disaccharide galactose-(? 1?3)-N-acetylgalactosamine, and occasionally for terminal ?-galactose and ?-N-acetylgalactosamine. Staining with three fucose-binding lectins revealed that the linkage between terminal ?-fucose residues to the constituent oligosaccharide chains varied between epithelium, blastema and stroma. One case in particular contained elements which exhibited certain morphologic and histochemical properties serving to identify them as neoplastic analogs of different regions of the normal nephron and collecting duct. Structures in this specimen identified as glomeruloid bodies showed a lectin binding capacity and immunohistochemical profile identical to normal glomeruli. In addition, three distinct populations of neoplastic tubules were present. One of these populations showed morphologic and histochemical properties common to normal proximal tubules, another to normal thin limbs, and a third to normal collecting ducts. Thus, lectin histochemistry provides a new and more accurate approach to characterizing the content and distribution of glycoconjugates in normal tissues and allows for more suitable comparisons with neoplastic counterparts.
